A doctoral work on the Effect of mismatch repair defects on the
response of colon carcinoma cells to chemotherapy
The response or nonresponse of a tumor to chemotherapy depends on the
composition of genetical lesions and the resulting functional
deficiencies in the individual tumors. Only 30% of colorectal
carcinomas respond to chemotherapy but the prediction of the responders
is still not possible.
We investigate the cellular response of tumor cells to chemotherapy
with the aim of understanding the molecular determinants of the
response. This might enable us to predict if and how a tumor would
react to a certain therapy. The present work concerns the influence of
the mismatch-repair defects on the response to the first-line
chemotherapeutic agent irinotecan.
An array of techniques of cell biology (cell culture, cytotoxicity
assays, determination of apoptosis, necrosis and senescence),
immunochemistry (immunoprecipitation, FACS analysis, immunofluorescent
labelling, western blots) and molecular biology (RNAi, RT-PCR, reporter
assays, adenovirus construction and possibly oligonucleotide arrays, )
as well as animal models will be used during the work. The previous
doctoral theses on that subject have been published in Int J. Cancer
(Magrini et al.2002) and in Oncogene (Bhonde et al. 2004, 2005).
The suitable candidate will have a solid diploma in biology or
biochemistry, a routine experience in basic techniques of molecular
biology and cell biology and a genuine interest for scientific work on
clinically relevant questions. This position is immediately available.
